January 20 – The Battle of Rio de Janeiro: Portuguese forces led by Estácio de Sá successfully expel the French from Rio de Janeiro.
January 23 – After a reign of 45 years, the Jiajing Emperor, Zhu Houcong, dies in the Forbidden City of China.
January – A Spanish force under Captain Juan Pardo establishes Fort San Juan in the Native American settlement of Joara, marking the first European settlement in present-day North Carolina.
February 4 – Prince Zhu Zaiji, son of the Jiajing Emperor, ascends the throne of the Ming Dynasty as the Longqing Emperor.
February 10 – Henry Stuart, Lord Darnley, husband of Mary, Queen of Scots, is murdered at the Provost’s House in Kirk o’ Field, Edinburgh.
March 13 – Battle of Oosterweel: A Spanish mercenary army ambushes and kills a group of rebels near Antwerp, marking the start of the Eighty Years’ War.

April 9 – The Battle of Thanesar occurs in present-day Haryana, India, where Mughal Emperor Akbar defeats over 7,000 warriors of the Sanyasi Hindu sect with only 300 men.
April 10 – Henrique I Nerika a Mpudi becomes the new ruler of the Kingdom of Kongo, succeeding his nephew, Bernardo I.
April 12 – The Earl of Bothwell is acquitted of murder charges in the killing of Lord Darnley and plans to marry Mary, Queen of Scots.
April 20 – The Ainslie Tavern Bond is signed, endorsing Bothwell as a suitable husband for Mary.
April 24 – Bothwell captures Mary at his castle in Dunbar and commits acts of violence against her.
